Research Associate in Multi-Omics Informatics
This post will be based in the group of Prof. Richard Unwin at the University of Manchester, which is based in the Division of Cancer Sciences. We are primarily interested in the application of mass spectrometry and proteomics to explore the mechanisms behind diseases of ageing.
This project is part of an ongoing programme of research to explore the molecular changes which underpin the risk of development of Age-Related Macular Degeneration in the human eye. AMD is the most common cause of adult sight loss in the developed world, and for most patients there is no effective therapy to reverse, or halt disease. Understanding the processes that contribute to its development in the early stages is key to solving this problem.
Funded by the Macular Society, this project aims to collect and integrate transcriptomics data and proteomics data acquired on human retinal tissues collected from individuals with high- and low- genetic risk of disease to define the molecular changes that define an environment in which AMD occurs. This has been successful in respect of the proteome in an analysis of the acellular Bruch’s membrane layer of the retina (DOI: 10.1073/pnas.2118510119), and transcriptomics data on samples of retinal pigment epithelial cells and neurosensory retina is ongoing. This project will generate matched proteomics datasets for samples with pre-existing transcriptomics data and will integrate these to answer key questions regarding changes in transcript and protein levels as a result if genetic disease risk at both chromosomes 1 and 10, including preliminary proteogenomic analysis to identify disease associated protein splice isoforms.
